Validation criteria (for users)
The validation score indicates how well the quality assurance data supports the specificity of the antibody towards the expected human target protein in an application-specific manner.
The antibody must be scored into one of three main validation categories:

  • Supportive
  • Uncertain
  • Not supportive

Image(s) showing application-specific results from validation scores supportive or uncertain must be submitted with a short text. Results (images) for non-supportive results do not need to be submitted, but a short text description is mandatory.

In the following, examples of text for the three main categories for four common research applications are listed, but more applications will be added in due course.

Validation scores: Immunohistochemistry

  • Supportive
    • Two independent antibodies targeting one protein yielding similar staining patterns. Staining pattern consistent with experimental and/or bioinformatics data.
    • Staining pattern consistent with experimental and/or bioinformatics data.
  • Uncertain
    • Staining pattern partly consistent with experimental and/or bioinformatics data.
    • No experimental and/or bioinformatics data available, but some degree of consistent staining is obtained.
  • Non-supportive
    • Staining pattern not consistent with experimental and/or bioinformatics data.

Validation scores: Immunofluorescence

  • Supportive
    • Two independent (paired) antibodies towards the same target showing the same staining.
    • One/multiple localizations supported by literature.
    • Multiple localizations partly supported (at least one) by literature.
    • One/multiple localizations in cytoplasm (i.e. golgi, mitochondria, ER etc) with literature supporting cytoplasmic localization.
  • Uncertain
    • One/multiple localizations where there is no literature available.
    • Not decisive. (One/multiple localizations where the literature is partly supporting and partly conflicting.)
    • Annotated weak/cytoplasmic/granular even if supported by literature for cytoplasmic localization.
  • Non-supportive
    • No staining.
    • Annotated weak/cytoplasmic/granular and there is no literature available.
    • Localization not consistent with literature.

Validation scores: Protein array

  • Supportive
    • Only binding to its own antigen in a background of many antigens.
  • Uncertain
    • The major binding is to its own antigen (less than 40% to all other antigens).
  • Non-supportive
    • No or weak signal.
    • Low specificity (one antigen with >40% signal or more than two antigens with signal >15%).

Validation scores: Western blot

  • Supportive
    • Two independent (paired) antibodies towards the same target showing the same band(s).
    • Single band corresponding to the predicted size in kDa (+/-20%).
    • Band of predicted size in kDa (+/-20%) with additional (less pronounced) bands present.
    • Single band larger than predicted size in kDa (+20%) but partly supported by predicted transmembrane region, signal peptide or by other available data.
  • Uncertain
    • No bands detected (but target protein probably not present in the lysate).
    • Target too small/large to be analyzed with the present setup.
  • Non-supportive
    • Single band differing more than +/-20% from predicted size in kDa and not supported by predicted transmembrane region, signal peptide or by other available data.
    • Weak band of predicted size but with additional bands of higher intensity also present.
    • Only bands not corresponding to the predicted size.
